Xanthine oxidase (XO, EC ) is present in appreciable amounts in the liver and jejunum in healthy individuals. However, in various liver disorders, XO is released into circulation. Therefore, determination of serum XO level serves as a sensitive indicator of acute liver damage such as jaundice. BioVision has developed an easy and sensitive assay to determine XO in variety of samples. In the assay, XO oxidizes xanthine to hydrogen peroxide (H₂O₂) which reacts stoichiometrically with OxiRed™ Probe to generate color (at λ = 570 nm) and fluorescence (at Ex/Em = 535/587 nm). Since the color or fluorescence intensity is proportional to XO content, the XO activity can be accurately measured. The kit detects 1-100 mU xanthine oxidase in 100 µl reaction volume.

How should blood be collected?
We recommend using heparin tubes.
Could you tell us the information about the each source of Enzyme Mix and XO Positive Control ?
K710 enzyme mix is from Horse Radish plant and the positive control is from buttermilk (food source).
I'd like to freeze my sample before use with this kit. Should I simply freeze the pellet, or is there a more appropriate step in the recommended sample preparation for this kit to freeze the samples?
The cell pellet after removing all media can be stored at -80C with a small volume of PBS. It is not ideal to store cells with medium. Once thawed, the cells can be washed once with PBS and then the PBS can be removed. At this step the assay buffer can be used for extraction according to the protocol.
Can we use frozen samples with this assay?
We would recommend using fresh samples. However, frozen samples can be used too provided they have not undergone multiple freeze thaw cycles.
Can the kit work on bacteria or yeast cells?
The kit has been standardized for mammalian cells only.
What is the exact volume of sample required for this assay?
There is no specific volume we can recommend for the amount any sample to be used since it is completely sample concentration and quality based. You have to do a pilot expt with multiple sample volumes to determine the optimal volume which gives a reading within the linear range of the standard curve. Can we use an alternate buffer for sample preparation (cell lysis, sample dilutions etc)?
Our assay buffers are optimized for the reactions they are designed for. They not only contain some detergents for efficient lysis of your cells/tissue, but also contain some proprietary components required for the further reactions. Therefore, we highly recommend using the buffers provided in the kit for the best results.
