The VisionBlue™ Quick Cell Proliferation Assay Kit provides by far the most sensitive and easiest means for quantifying cell proliferation and viability. Simply add the single reagent to cell culture, incubate and read the fluorescence intensity. The VisionBlueTM assay utilizes the redox dye resazurin which is not fluorescent, but upon reduction by viable metabolically active cells, the dye becomes highly fluorescent (Ex = 530 - 570 nm; Em = 590-620 nm). Therefore, viable metabolically active cells can be easily measured.

 Can you incubate cells with Visionblue reagent overnight?
Yes. However, signals from higher cell density samples may have “saturated,” which means the linearity of reagent may have reached a plateau. If this occurs, decrease the incubation time.
I can see a difference in the density of my cells, why doesn’t the assay reflect the difference?
Check to make sure your cell densities are in the linear range of the assay. Too few cells may fall below the limit of detection, and too many cells may saturate the OD. Perform an initial experiment to check the best linear range for your cell type and experimental set-up. You may need to vary the cell density assay incubation time.
What is the advantage of resazurin over MTT/XTT/WST-1 reagents?
MTT/WST-1/XTT are coloreimetric based reagents whereas Resazurin could be both colorimetric or fluorometric. Unlike MTT, Resazurin do not require cell lysis, thereby allowing kinetic monitoring of the same samples at different timepoints.
Why do I see a high background?
The reagent has been exposed to light for extended periods. It is important to store the reagent in a dark place protected from light. Serum and albumin in media may lead to false positives. It is best to use medium containing less than 10% added protein.
Can the kit work on bacteria or yeast cells?
The kit has been standardized for mammalian cells only
What is the exact volume of sample required for this assay?
There is no specific volume we can recommend for the amount any sample to be used since it is completely sample concentration and quality based. You have to do a pilot expt with multiple sample volumes to determine the optimal volume which gives a reading within the linear range of the standard curve. Please refer to the citations for this product to see what other clients have used with similar sample types.

What is the shelf life of this kit?
 BioVisions’s assay kits expire 6 months from the date of shipment. Some components of the kit may expire sooner as mentioned in the data sheet

Can we use an alternate buffer for sample preparation (cell lysis, sample dilutions etc)?
Our assay buffers are optimized for the reactions they are designed for. They not only contain some detergents for efficient lysis of your cells/tissue, but also contain some proprietary components required for the further reactions. Therefore, we highly recommend using the buffers provided in the kit for the best results.
