Details

Uric acid in serum is the end product of purine metabolism, and is cleared through the kidney by glomerular filtration. However, human often lacks the necessary enzyme called urate oxidase (Uricase), and therefore abnormal uric acid may be accumulated in blood. Recent evidences show the close association between serum urate level and cardiovascular morbidity and mortality, especially among persons at high cardiovascular risk, including those with hypertension, diabetes and congestion heart failure. BioVision’s Uric Acid Assay Kit provides a convenient means for detecting uric acid in biological samples such as serum and urine. Pretrea™ent of samples are not required. Uric acid level can be measured using fluorometric (at Ex/Em = 535/587 nm) or colorimetric (at λ= 570 nm) methods.

Can this kit be used with cells/tissues?
Yes, it is possible to use this kit with cells or tissues. The assay buffer can be used to lyse 1-2 million cells or 10-100mg tissue per assay. It is recommended to run a pilit assay with different amounts of the sample such that the readings are within the linear range of the standard curve.
What kind of sample preparation is needed for urine?
Urine samples can be used directly after spinning down to remove any suspended particulate matter. Different volumes should be tested to optimize the amount required for readings within the linear range of the std. curve.
The OD values are very low. What could be the problem?
The most common reason for low OD values is that the Assay buffer was cold, which led to a slow reaction.
What is the principle behind using the colorimetric versus the fluorometric assay?
The fluorometric assay is atleast 10 times more sensitive than the colorimetric assay. For 0-5 nmol of Uric acid the Flurometric method can be used. For >5 up to 40nmol of Uric acid the colorimetric method can be used.
What is the exact volume of sample required for this assay?
There is no specific volume we can recommend for the amount any sample to be used since it is completely sample concentration and quality based. It is recommended to do a pilot expt with multiple sample volumes to determine the optimal volume which gives a reading within the linear range of the standard curve. Please refer to the citations for this product to see what other clients have used with similar sample types.
Can frozen samples be used with this assay?
Fresh samples are always preferred over frozen samples. However, frozen samples can also be used, provided, they were frozen right after isolation, were not freeze thawed multiple time (for which we recommend aliquoting the samples before freezing) and have been frozen for relatively short periods.
How are samples normalized against protein concentration?
A detergent-compatible protein quantitation assay can be used with the supernatants from cell/tissue lysates or with any other liquid sample in the assay buffer.
Can individual components of this kit be purchased separately?
Yes, any of the kit's components can be purchased separately without having to buy the whole kit. Please refer to the component Cat #s mentioned on the datasheet for ordering.
The standard curve shows high values throughout and is flat. What could be the problem?
The most common reason for a flat std. curve is if the probe got oxidized. The probe should be light pinkish in color and if it already pink or red, this means due to exposure to light/misture and/or improper storage, the probe got oxidized.
Can white opaque plates be used for the fluorometric assay?
Yes, but only for a top-reading fluorometer. Transparent bottom black plates should be used for fluorometers which have top-reading systems.
What is the shelf life of this kit?
This kit is good for 12 months from the date of shipment in the unopened form when stored at the appropriate temperature and appropriate conditions. After opening and reconstitution, some of the components in this kit are good for 2 months at -20°C. Please refer to the datasheet for storage information and shelf life of each of the components.
