Urea is a waste product which is produced in the liver, dissolved and transported in the blood (in a concentration of 1.6-7.5 mM), and excreted by the kidney. Urea plays a very important role in protein catabolism, removal of toxic ammonia from the body, and the countercurrent system which allows for reabsorption of water and critical ions in the nephrons. Urea concentration is an important indicator for the medical clinicians to assess function of the kidney and other organs in patients. BioVision’s Urea Assay Kit II is based on Jung’s method with a modification that delivers more robust and sensitive data. The urea is condensed with o-phthalaldehyde (OPA), followed by a reaction to form a colored product with strong absorbance at 505 nm. This assay kit is fast, sensitive & easy to use. It can measure less than 10 μM urea in 96-well plate assay.

Can you test for urea in the cell culture medium?
Cells do not normally secrete urea into the medium. So we do not expect to detect urea easily in this sample type. Any interference from FBS can always be subtracted from the sample readings if needed as long as there is enough urea in the medium volume used per well. We have not tested culture media with this kit. The media, if containing phenol red will likely change color due to the pH change during the assay and this can contribute to color.
What is the difference between K375 and K376?
Colorimteric Kit II (K376) has a different principle than K375. K376 is a more robust and sensitive kit (can measure lower than 10uM). The std curve in K376 shows that it is linear over a broader range than K375. You can create a std curve with 1-5 nmoles/well using K376 as well if your samples are in that low range. K376: BioVision’s Urea Assay Kit II is based on Jung’s method with a modification that delivers more robust and sensitive data. The urea is condensed with o-phthalaldehyde (OPA), followed by a reaction to form a colored product with strong absorbance at 505 nm. This assay kit is fast, sensitive & easy to use. It can measure less than 10 µM urea in 96-well plate assay. K375: Urea reacts as substrate with compounds in the presence of enzymes to form a product that reacts with the OxiRed probe to generate color (?max=570nm). The optical density of produced color has a direct relationship with Urea concentration in the solution. The kit can detect as low as 0.5 nmol per well or 10 µM Urea. Ammonia can create background in this assay.
Can you adapt the kit for 384 well plate?
You could scale down the assay into a 384-well format, making sure the relative proportions of the components in the reaction per well remains the same as in the 96-well format. Scaling down might require optimization (it is difficult to predict reaction completion time and concentration of reactants in very small volumes) to get best results. We have not tested the kit using a 384-well format and have not received reports from other customer about it yet since the improved kit II is relatively new.
Can the kit work on bacteria or yeast cells?
The kit has been standardized for mammalian cells only
What is the exact volume of sample required for this assay?
There is no specific volume we can recommend for the amount any sample to be used since it is completely sample concentration and quality based. You have to do a pilot expt with multiple sample volumes to determine the optimal volume which gives a reading within the linear range of the standard curve. Please refer to the citations for this product to see what other clients have used with similar sample types.
Do you have trial sizes of this kit?

What is the shelf life of this kit?
 BioVisions’s assay kits expire 6 months from the date of shipment. Some components of the kit may expire sooner as mentioned in the data sheet

Can we use an alternate buffer for sample preparation (cell lysis, sample dilutions etc)?
Our assay buffers are optimized for the reactions they are designed for. They not only contain some detergents for efficient lysis of your cells/tissue, but also contain some proprietary components required for the further reactions. Therefore, we highly recommend using the buffers provided in the kit for the best results.
