Details

Thioredoxin reductase (TrxR) (EC 1.8.1.9) is a ubiquitous enzyme which is involved in many cellular processes such as cell growth, p53 activity, and protection against oxidation stress, etc. The mammalian TrxR reduces thioredoxins as well as non-disulfide substrates such as selenite, lipoic acids, lipid hydroperoxides, and hydrogen peroxide. BioVision’s Thioredoxin Reductase Assay Kit provides a convenient colorimetric assay for detecting TrxR activity in various samples. In the assay TrxR catalyzes the reduction of 5, 5’-dithiobis (2-nitrobenzoic) acid (DTNB) with NADPH to 5-thio-2-nitrobenzoic acid (TNB2-), which generate a strong yellow color (λmax = 412 nm). Since in crude biological samples other enzymes, such as glutathione reductase and glutathione peroxidase, can also reduce DTNB, therefore, TrxR specific inhibitor is utilized to determine TrxR specific activity. Two assays are performed: the first measurement is of the total DTNB reduction by the sample, and the second one is the DTNB reduction by the sample in the presence of the TrxR specific inhibitor. The difference between the two results is the DTNB reduction by TrxR.

What is the source of the positive control?
The positive control is thioredoxin reductase isolated from Rat liver.
Do you know how this molecule can be localised in the plasma/serum/cell supernatant? Is it released from dying/dead cells? Is this actively secreted outside of the cells?
Thioredoxin reductase 1 is a cytoplasmic enzyme. K763 is not testing any receptor but the activity of the enzyme. If the cell membrane is damaged or pinched off this can be released into medium. Isoform 1 induces actin and tubulin polymerization, leading to formation of cell membrane protrusions.Thioredoxin reductase 2 is localized in the mitochondria and expressed in prostate, testis, liver. Our kit is not specific for any isoform. K763 measures the total TrxR activity in the sample. If the customer is using mitochondrial prep from prostrate, liver or testes, they will measure TrxR2 activity. So it depends on the sample.
Can we use frozen samples with this assay?
We would recommend using fresh samples. However, frozen samples can be used too provided they have not undergone multiple freeze thaw cycles.
Can the kit work on bacteria or yeast cells?
The kit has been standardized for mammalian cells only.
What is the exact volume of sample required for this assay?
There is no specific volume we can recommend for the amount any sample to be used since it is completely sample concentration and quality based. You have to do a pilot expt with multiple sample volumes to determine the optimal volume which gives a reading within the linear range of the standard curve. Please refer to the citations for this product to see what other clients have used with similar sample types.
Do you have trial sizes of this kit?
Unfortunately, we do not have trial sizes of this kit available. However, if you are based in the US or Canada, we will give you a 10% off list price introductory discount on its purchase price. If you are based out of this area please contact your regional BioVision distributor.
What is the shelf life of this kit?
BioVisions’s assay kits expire 6-12 months from the date of shipment. Some components of the kit may expire sooner as mentioned in the data sheet.
Can we purchase individual components of this kit?
Yes, you can purchase any of the kit's components without the whole kit. Please refer to the component Cat #s mentioned on the datasheet for ordering.
Can we use an alternate buffer for sample preparation (cell lysis, sample dilutions etc)?
Our assay buffers are optimized for the reactions they are designed for. They not only contain some detergents for efficient lysis of your cells/tissue, but also contain some proprietary components required for the further reactions. Therefore, we highly recommend using the buffers provided in the kit for the best results.
