Protein-Protein Interaction (PPI) Inhibitors.

Proteins play a key role in major biological processes both inside and outside the cell. Few proteins function independently while many others interact with one or more proteins forming protein-protein complexes. Some of the biological effects of these protein-protein interactions (PPIs) are alteration of enzyme properties, substrate channeling, creating new binding sites, destroying the protein, changes in specificity of the protein, changes in upstream or downstream events etc. This creates dysregulation in normal cellular processes and may lead to many diseases and complications.
Small molecule PPI inhibitors like Tirofiban, Cyclosporine and many more have inspired confidence in this area of research. These small molecule inhibitors modulate PPIs via binding to the so called “hot spot” residues at the PPI interface. These PPI inhibitors seem to have affinities similar to drug leads. Hence, PPI inhibitors represent an attractive target for therapeutic interventions and are becoming increasingly important in drug discovery.
