PI3K-mTOR Inhibitors

Phosphatidyl Inositol 3 kinases (PI3-Ks) are a family of Serine/Threonine phosphorylases which phosphorylate the 3rd position hydroxyl group of the inositol ring of phosphatidylinositol. Based on structure, regulation and substrate specificity, the PI3K family is divided into three different classes: Class I, II and III. Due to the versatility of the end product (various 3-P-inositides like PI3P) in various signal transduction networks, PI3K enzymes function in a diverse array of cellular processes like growth, division, differentiation, migration, survival and overall modulation of responses to different environmental cues like nutrient deprivation, stress, infection, inflammation etc. PI3Ks also function in intracellular trafficking of proteins. Through interaction with the Insulin receptor, they modulate glucose uptake into a cell. This is the basis of their involvement in Diabetes. Majority of the downstream effects are brought about through the ability of PI3Ks to activate Protein Kinase B or Akt (PI3K-mTOR pathway for example). Mutations in PI3K enzymes are found in different cancers. PI3K has also been implicated in long-term potentiation underlying synaptic plasticity, learning and memory. PI3K inhibitors constitute an important class of modulators of a large number of cellular processes. Wortmannin (irreversible) and LY294002 (reversible) are the two most well-known inhibitors of PI3K enzymes. Many PI3K inhibitors are in different phases of clinical trials for the treatment of cancer, neurodegenerative diseases and respiratory inflammatory diseases to name a few.
