Peroxisome proliferator-activated receptor-γ (PPARG) is a type II nuclear hormone receptor that belongs to the family of Peroxisome proliferator-activated receptors. This ligand-activated transcription factor is mainly present in the liver, macrophages, adipose tissue, and colon. PPARG exists in 2 isoforms in humans and mice: PPARG1 (all tissues except muscle) and PPARG2 (in adipose tissue and intestine). PPARG forms heterodimers with retinoid X receptor (RXR), binds to the consensus DR1 sequence in the promoters of target genes, and regulates their transcriptional activity. PPARG is known to promote differentiation and survival of adipocytes, lipogenesis, and regulate insulin sensitivity. PPARG also plays a critical role in the circadian rhythms. Circadian expression of PPARG has been reported in mouse liver, blood vessels, and adipose tissue. PPARG also regulates circadian clock components such as Bmal1 and Rorg. Deletion of PPARG dampens or abrogates these rhythms and disturbs metabolic and energy homeostasis. PPARG is implicated in the pathogenesis of several diseases such as diabetes, atherosclerosis, obesity, and cancer. Therefore PPARG could serve as a promising target to treat metabolic diseases. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. HRP-Streptavidin is added to the plate, and post-incubation any unattached conjugates are washed off by Wash Buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of PPARG in the sample or standard.
