Plasmin (EC is a serine protease occurring in plasma as plasminogen. Upon activation via cleavage by plasminogen activators; plasmin solubilizes fibrin clots and activates and/or degrades compounds of the coagulation and complement systems. Plasminogen-Plasmin system has also been implicated in a wide variety of physiologic and pathologic processes, including tumor growth, invasion and metastasis. BioVision's Plasmin activity assay kit utilizes the ability of Plasmin to proteolytically cleave a synthetic substrate and release a fluorophore, AMC, which can be easily quantified by fluorescence microplate readers. This assay kit is simple, rapid and can detect Plasmin activity as low as 10 ng in a variety of samples.

Does the kit measure the plasmin activity, which is already available in the sample or is there any activator which induces the conversion of plasminogen to plasmin?
The kit measures Plasmin that is already present in the sample. No activators are included in the kit.
For the plasmin assay, looks like you can measure plasminogen too with addition of urokinase, #7696 to assay buffer.What concentration of urokinase do you use in this assay buffer to measure plasminogen?
We used 0.5-1 ug urokinase and incubated at 37C for 1 h. Typically the Urokinase:plasminogen ratio is 1:5.
Can the kit work on bacteria or yeast cells?
The kit has been standardized for mammalian cells only
What is the exact volume of sample required for this assay?
There is no specific volume we can recommend for the amount any sample to be used since it is completely sample concentration and quality based. You have to do a pilot expt with multiple sample volumes to determine the optimal volume which gives a reading within the linear range of the standard curve. Please refer to the citations for this product to see what other clients have used with similar sample types.

Can we use an alternate buffer for sample preparation (cell lysis, sample dilutions etc)?
Our assay buffers are optimized for the reactions they are designed for. They not only contain some detergents for efficient lysis of your cells/tissue, but also contain some proprietary components required for the further reactions. Therefore, we highly recommend using the buffers provided in the kit for the best results.
