Details

Triglycerides (TG) are the main constituents of vegetable oil, animal fat, LDL and VLDL, and play an important role as transporters of fatty acids as well as an energy source. TG is broken down into fatty acids and glycerol, after which both can serve as substrates for energy producing and metabolic pathways. High blood levels of TG are implicated in atherosclerosis, heart disease and stroke as well as in pancreatitis. Biovision’s PicoProbe™ Triglyceride Assay kit is suitable for measuring triglyceride levels in samples, which contain reducing substances that may interfere with oxidase-based assays. In this assay, TG is hydrolyzed to glycerol and fatty acid. The glycerol reacts with Triglyceride Enzyme Mix to form an intermediate product, which in turn reacts with PicoProbe & Developer to generate the fluorescence. The generated fluorescence is directly proportional to the amount of triglycerides. This high-throughput adaptable assay kit is simple, sensitive and easy to use & can detect less than 0.4 µM triglycerides in a variety of samples.

What is the difference between K614 and K622 (Triglyceride colorimetric/fluorometric assay kit)?
The picoprobe allows K614 to be manifold more sensitive than the K622. K614 detects triglycerides <0.4uM. K622 can detect 2uM-10mM triglycerides.
The values of the std curve are in the low hundreds. Why?
It is important to warm up the probe to >20C and the assay bufefr to RT to ensure that the reaction proceeds according to the normal time of incubation. Incompletely thawed or cold reagents can lead to a slow reaction and low values.
Can this asay be used for a HTS?
Yes, this assay is HTS adaptable.
Is deproteinizing recommended for all samples?
Deproteinization is not required for this assay.
What can cause background in this assay?
Glycerol and NADH are the primary sources of background in this assay. Substraction of background will help correct for this contribution. BSA (1% or higher) and PMSF might also interfere with the assay.
Will di- and mono- acyl glycerols be detected by this assay?
The Lipase enzyme that hydrolyzes TG is specific for only Triacylglycerol and thus di- and mono-glycerides will not be recognized in this assay.
Can tissue be stored in PBS-glycerol and then used to homogenize for the assay?
Glycerol creates background and hence tissu/cell storage in glycerol must be avoided, or the glycerol will need to be washed off before homogenization.
How are samples normalized against protein concentration?
A protein quantitation assay can be used with the supernatants from cell/tissue lysates or with any other liquid sample in the assay buffer.
Are trial sizes of this kit available?
Unfortunately, we do not have trial sizes of this kit available. However, for customers based in the US or Canada, a 10% off list price introductory discount can be offered on the list price. For all other customers based out of this area our regional BioVision distributor should be contacted
Can individual components of this kit be purchased separately?
Yes, any of the kit's components can be purchased separately without having to buy the whole kit. Please refer to the component Cat #s mentioned on the datasheet for ordering.
Can alternate buffers be used for sample preparation (cell lysis, sample dilutions etc)?
Our assay buffers are optimized for the reactions they are designed for. They not only contain some detergents for efficient lysis of your cells/tissue, but also contain some proprietary components required for the further reactions. Therefore, we highly recommend using the buffers provided in the kit for the best results.
