NADP (nicotinamide dinucleotide) is a major electron donor for anabolic reactions such as lipid and nucleic acid synthesis. The reduced form, NADPH is a cofactor for many redox enzymes. It is of increasing interest to be able to measure low level of intracellular NADPH. BioVision’s PicoProbe™ NADPH Quantitation Assay kit provides a highly sensitive method to detect intracellular NADPH. NADH is not detected as the NADPH Recycling Enzyme Mix specifically recognizes NADPH in the enzyme recycling reaction. The assay is rapid, simple and does not require purifying NADPH from samples. The kit can measure less than 40 nM NADPH in a variety of samples.

What is the minimum number of suspension cells I can use for this kit to detect?
Typically 1 million cells per sample is a good starting point. The #cells depends on the amount of NADPH in the cell at the time of the assay. The assay detects between 1-10 pmoles of NADPH per well.
What is the nature of the PicoProbe? Is that a NADH/NADPH Developer as in kits K337 and K347?
The picoprobe in K338 /K349 is a chemical dye that is more sensitive than the developer in K337 or K347.
Can the kit work on bacteria or yeast cells?
The kit has been standardized for mammalian cells only
What is the exact volume of sample required for this assay?
There is no specific volume we can recommend for the amount any sample to be used since it is completely sample concentration and quality based. You have to do a pilot expt with multiple sample volumes to determine the optimal volume which gives a reading within the linear range of the standard curve. What is the shelf life of this kit?
 BioVisions’s assay kits expire 6 months from the date of shipment. Some components of the kit may expire sooner as mentioned in the data sheet

Can we use an alternate buffer for sample preparation (cell lysis, sample dilutions etc)?
Our assay buffers are optimized for the reactions they are designed for. They not only contain some detergents for efficient lysis of your cells/tissue, but also contain some proprietary components required for the further reactions. Therefore, we highly recommend using the buffers provided in the kit for the best results.
