Lactate Dehydrogenase (LDH, EC is a stable enzyme and is present in all cell types. Cell damage leads to release of LDH enzyme into the media and its activity is widely used as a marker for cytotoxicity. It reversibly converts lactate into pyruvate, with the concomitant interconversion of NADH and NAD+. BioVision’s PicoProbe™ LDH-Cytotoxicity Fluorometric Assay Kit provides a sensitive, quick, and easy way for detection of LDH released from damaged cells. In this assay, LDH converts lactate to pyruvate and NADH, which reduces a proprietary probe to an intensely fluorescent product (Ex/Em = 535/587 nm). The amount of fluorescence is directly proportional to the number of damaged cells. The assay is adaptable to high-throughput format and can be completed in less than 20 min. Sensitivity: ~ 100 cells.

We would like to know if this kit can be used for supernatant samples that have been frozen?
It is best to use this with fresh samples rather than frozen. The protocol will have to be changed slightly to accommodate the frozen samples. Also, the success of the assay would depend on numerous extra criteria including how well the samples were frozen, how long they were frozen, number of times they were thawed out etc, which increases the complexity. Therefore it is recommend to stick with fresh samples.
Does serum affects the assay?
Serum will increase the OD reading (0.5). However, if your background, control media and testing media all contain the same amount of serum, the affect might be ignored (depending on each situation), since it will be subtracted from your testing samples.
Can the kit work on bacteria or yeast cells?
The kit has been standardized for mammalian cells only
What is the exact volume of sample required for this assay?
There is no specific volume we can recommend for the amount any sample to be used since it is completely sample concentration and quality based. You have to do a pilot expt with multiple sample volumes to determine the optimal volume which gives a reading within the linear range of the standard curve. What is the shelf life of this kit?

Can we use an alternate buffer for sample preparation (cell lysis, sample dilutions etc)?
Our assay buffers are optimized for the reactions they are designed for. They not only contain some detergents for efficient lysis of your cells/tissue, but also contain some proprietary components required for the further reactions. Therefore, we highly recommend using the buffers provided in the kit for the best results.
