P-cadherin also known as Cadherin-3 or Placental cadherin, is a classical cell-to-cell adhesion molecule with a homeostatic function in several normal tissues. Cadherins are calcium-dependent cell adhesion proteins. They preferentially interact with themselves in a homophilic manner in connecting cells. Thus, cadherins contributes to the sorting of heterogeneous cell types. The cadherin family is essential in maintaining the cell-cell contact and regulate cytoskeletal complexes. The cadherin superfamily includes cadherins, protocadherins, desmogleins, desmocollins, etc. P-cadherin acts as a tumor suppressor, since its absence is associated with a more aggressive cancer cell phenotype. The overexpression of this molecule is linked to significant tumor promoting effects in the breast, ovarian, prostate, endometrial, skin, gastric, pancreas and colon neoplasms. P-cadherin acts as a pro-adhesive and anti-invasive/antimigratory molecule in colon carcinoma cells and stable knockdown in RT-112 cells diminishes invasion and migration, and thus promotes intercellular adhesion. BioVision’s P-Cadherin (Human) ELISA Kit is used for the quantitative detection of P-Cadherin in serum, plasma, tissue homogenates and other biological fluids. It is based on the principle of sandwich ELISA. The capture antibody is pre-coated on 96-well plates. The standards, test samples and biotin conjugated detection antibody are added to the wells subsequently, and washed with wash buffer. HRP-Streptavidin is added and unbound conjugates are washed away with wash buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of P-Cadherin in the sample.
