Osterix (OSX, SP7) is a zinc-finger-containing transcription factor that belongs to the family of specificity proteins (Sp). During the development stage in bones, OSX is specifically expressed in osteocytes and osteoblasts. It is a master regulator of osteoblast differentiation and bone formation during embryonic development and adulthood. BMP2 induces Osx gene expression via Dlx5. Osx then activates osteoblastic genes such as Col1A1, Osteopontin, Osteocalcin, and Bone sialoprotein, all of which are required for osteoblast formation. Other factors such as TNF-α, Parathyroid hormone (PTH), and p53 negatively regulate Osx expression. Absence or knockdown of OSX results in impairment of osteoblastic activity and maturation of osteoclasts. On the other hand, increased expression contributes to bone metastasis in breast cancer patients. Mutations in OSX are associated with Osteogenesis imperfecta (OI), a skeletal disorder associated with low bone mass and bone mineral density with higher susceptibility to bone fractures. Since it is a classic osteoblast marker, OSX could potentially be used to treat bone fractures and injuries. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. HRP-Streptavidin is added to the plate, and post-incubation any unattached conjugates are washed off by Wash Buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of OSX in the sample or standard.
