Neutrophil Elastase (NE, EC 3.4.21.37, leukocyte elastase, elastase 2, or serine elastase) is a cytotoxic serine protease. It is stored in the azurophil granules of neutrophil granulocyte and is released following cell stimulation, e.g. by pathogens, immune complexes or chemotactic agents (PMA). When the extracellular NE concentration exceeds the buffering capacity of endogenous inhibitors, it causes degradation of a wide range of extracellular matrix proteins, including fibronectin, laminin, proteoglycans, collagens, and elastin. Extracellular elastase is implicated in the signs, symptoms and disease progression of inflammatory lung disorders (such as cystic fibrosis, COPD, lung emphysema) via its role in the inflammatory processes, mucus overproduction and lung tissue damage. BioVision's Neutrophil Elastase Activity Assay Kit utilizes the ability of NE to proteolytically cleave a synthetic substrate and release a fluorophore, AFC, which can be easily quantified by fluorescence microplate readers. This assay kit is simple, rapid and can detect as low as 1 ng of Neutrophil Elastase in a variety of samples.

What's NE Enzyme Standard derived from (e.g. human NE)?
The standard is Elastase derived from human neutrophil
Is this kit specific only for neutrophil elastase or if it is also able to detect pancreatic elastase?
We have not tested the kit with pancreatic elastase. However, it is known that pancreatic elastase and neutrophil elastase a quite different sequence-wise and might not function the exact same way under similar conditions: http://www.ncbi.nlm.nih.gov/pmc/articles/PMC304622/
. Do you know if I can use your assay kit with my sputum sample ?
Neutrophil elastase assay (http://www.biovision.com/neutrophil-elastase-activity-assay-kit-fluorometric-9581.html) has been tested with blood samples because the enzyme is present in high concentrations in neutrophils. We have not tested sputum samples with this kit. As long as the sample you intend to use has active elastase enzyme, the sample type can work. The sample would need to be diluted to make into a non-viscous aqueous solution and the amount would need to be optimized such that the values are within the linear range of the std. curve.
Can the kit work on bacteria or yeast cells?
The kit has been standardized for mammalian cells only
What is the exact volume of sample required for this assay?
There is no specific volume we can recommend for the amount any sample to be used since it is completely sample concentration and quality based. You have to do a pilot expt with multiple sample volumes to determine the optimal volume which gives a reading within the linear range of the standard curve. Please refer to the citations for this product to see what other clients have used with similar sample types.

 BioVisions’s assay kits expire 6 months from the date of shipment. Some components of the kit may expire sooner as mentioned in the data sheet

Can we use an alternate buffer for sample preparation (cell lysis, sample dilutions etc)?
Our assay buffers are optimized for the reactions they are designed for. They not only contain some detergents for efficient lysis of your cells/tissue, but also contain some proprietary components required for the further reactions. Therefore, we highly recommend using the buffers provided in the kit for the best results.
