N-cadherin also known as Cadherin-2, CDH2 or Neural Cadherin, is a calcium-dependent single-chain transmembrane glycoprotein that mediates homotypic and heterotypic cell-cell adhesion. Cadherins are calcium-dependent cell adhesion proteins. They preferentially interact with themselves in a homophilic manner in connecting cells. Thus, cadherins contributes to the sorting of heterogeneous cell types. The cadherin family is essential in maintaining the cell-cell contact and regulate cytoskeletal complexes. In cardiac muscle, N-cadherin is an integral component in adherens junctions residing at intercalated discs, which function to mechanically and electrically couple adjacent cardiomyocytes. N-cadherin plays a role in development as a calcium dependent cell–cell adhesion glycoprotein that functions during gastrulation and is required for establishment of left-right asymmetry. Aberrant expression of N-cadherin is closely related to aspects of malignant tumor progression in humans, such as transformation, adhesion, apoptosis, angiogenesis, invasion and metastasis. Thus, N-cadherin can be a therapeutic target for tumor invasion and metastasis. BioVision’s N-Cadherin (Human) ELISA Kit is used for the quantitative detection of N-Cadherin in serum, plasma, tissue homogenates and other biological fluids. It is based on the principle of sandwich ELISA. The capture antibody is pre-coated on 96-well plates. The standards, test samples and biotin conjugated detection antibody are added to the wells subsequently, and washed with wash buffer. HRP-Streptavidin is added and unbound conjugates are washed away with wash buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of N-Cadherin in the sample.
