Histidine is one of the 20 naturally-occuring proteogenic amino acids and an essential dietary amino acid. The unique imidazole moiety of histidine has aromatic properties and a pKa ~ 6, making it a chemically versatile residue in proteins. In enzymes, histidine residues often play a critical role in the catalytic reaction mechanism, due to histidine’s ability to act as an acid or a base at physiological pH. The imidazole nitrogen atoms also make histidine a good chelator of metal ions such as zinc, iron and copper thereby making it an indispensable structural component in metalloproteins. Histidine is also required for the synthesis of histamine, a neurotransmitter and immunological signaling molecule. Histamine also acts as a paracrine hormone in the gut, promoting gastric acid secretion and digestive regulation. Dietary histidine deficiency results in anemia, fatigue and skin lesions. BioVision’s Histidine Assay Kit allows the quantification of L-Histidine levels in various biological samples. The assay is based on the selective enzymatic decarboxylation of histidine to histamine and subsequent metabolism of histamine, yielding an oxidized intermediate which reacts with a fluorogenic probe to form a stable fluorophore measured at Ex/Em = 538/587 nm. The assay is highly sensitive and is high-throughput adaptable and is not affected by the physiological concentrations of other proteogenic amino acids. The kit can detect down to 10 μM histidine in biological samples.
