Inhibition of histone deacetylase (HDAC) has been implicated to modulate transcription and to induce apoptosis or differentiation in cancer cells. However, screening of compounds for HDAC inhibition has been difficult due to the lack of convenient tools for analyzing HDAC activity. The new HDAC Inhibitor Drug Screening Kit provides a fast, fluorescence-based method that eliminates radioactivity, extractions, or chromatography, as used in traditional assays. The new procedure requires only two easy steps, both performed on the same microtiter plate. First, your inhibitor candidates are mixed with HeLa Nuclear Extract and HDAC fluorometric substrate, which comprises an acetylated lysine side chain. Deacetylation of the substrate sensitizes the substrate, so that, in the second step, treatment with the Lysine Developer produces a fluorophore. The fluorophore can be easily analyzed using a fluorescence plate reader or a fluorometer. The assay is well suited for high throughput screening applications.

Can we use this kit for measuring each HDAC1-11 separately, if we use HDAC-11 enzyme instead of HeLa Nuclear Extract? Is the substrate compatible with all of HDACs?
The substrate used in K340 is acetylated at the lysine moieties and deacetylated by HDACs. Yes, if you use individual HDACs instead of the HeLa extract in K340, then you can attribute the deacetylation activity to that of the specific HDAC. The substrate we use with K340 works with Class I and Class II b HDACs and this includes HDACs 1,2,3,6,8 and 10 and this information is based on the available research. We have not done any specific studies with individual HDACs in our lab and hence do not have any data from our side. But going by the literature, the substrate used should work for Class I and Class II b HDACs.

Why HeLa nuclear extract is used for screening HDAC activity?
The Hela Cell nuclear extract is used since it is an enriched source of HDAC activity. Choi K-C et al (2009) Cancer Res.; 69: 583 - 592.
Why do we recommend a high concentration of TSA?
. TSA is a reversible inhibitor. Hence excess inhibitor is added to ensure that the HDACs are completely inhibited. The IC50 varies for different HDACs and we intend to cover the whole range which is why this high concentration is used.
Can the kit work with all HDACs?
This kit could work with any HDAC enzyme. It is not specific for one particular HDAC.
Can the kit work on bacteria or yeast cells?
The kit has been standardized for mammalian cells only
What is the exact volume of sample required for this assay?
There is no specific volume we can recommend for the amount any sample to be used since it is completely sample concentration and quality based. You have to do a pilot expt with multiple sample volumes to determine the optimal volume which gives a reading within the linear range of the standard curve. Please refer to the citations for this product to see what other clients have used with similar sample types.

Can we use an alternate buffer for sample preparation (cell lysis, sample dilutions etc)?
Our assay buffers are optimized for the reactions they are designed for. They not only contain some detergents for efficient lysis of your cells/tissue, but also contain some proprietary components required for the further reactions. Therefore, we highly recommend using the buffers provided in the kit for the best results.
