FGF5 also known as Fibroblast growth factor 5 belongs to the fibroblast growth factor (FGF) family, which are glycosaminoglycan binding proteins which possess broad mitogenic and cell survival activities. It plays an important role in the regulation of embryonic development, cell proliferation, and cell differentiation. FGF family members possess broad mitogenic and cell survival activities. FGF5 is produced in the outer root sheath of the hair follicle as well as perifollicular macrophages, with maximum expression occurring in the late anagen phase of the hair cycle. The receptor for FGF5, FGFR1, is largely expressed in the dermal papilla cells of the hair follicle. FGF5 performs a critical role in the hair cycle, where it acts as the key signaling molecule in initiating the transition from the anagen (growth) phase to the catagen (regression) phase. FGF5 functions as an inhibitor of hair elongation by promoting progression from anagen, the growth phase of the hair follicle, into catagen the apoptosis-induced regression phase. Individuals with mutations in FGF5 exhibit familial trichomegaly, a condition that involves a significant increase in the portion of anagen phase hair as well as extremely long eyelashes. BioVision’s FGF5 (Human) ELISA Kit is used for the quantitative detection of FGF5 in serum, plasma, tissue homogenates and other biological fluids. It is based on the principle of sandwich ELISA. The capture antibody is pre-coated on 96-well plates. The standards, test samples and biotin conjugated detection antibody are added to the wells subsequently, and washed with wash buffer. HRP-Streptavidin is added and unbound conjugates are washed away with wash buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of FGF5 in the sample.
