FGF23 also known as Fibroblast growth factor 23 belongs to the fibroblast growth factor (FGF) family, which are glycosaminoglycan binding proteins which possess broad mitogenic and cell survival activities. It plays an important role in the regulation of embryonic development, cell proliferation, and cell differentiation. FGF family members possess broad mitogenic and cell survival activities. FGF-23 is a 32 kDa protein that is a key regulator of phosphorus and vitamin D metabolism. FGF23 is secreted by osteocytes in response to increased calcitriol. FGF23 acts on the kidneys, where it decreases the expression of NPT2, a sodium-phosphate cotransporter in the proximal tubule. The main function of FGF23 seems to be regulation of phosphate concentration in plasma and decreases the expression of NPT2, a sodium-phosphate cotransporter in the proximal tubule. Loss of FGF23 activity leads to increased phosphate levels and the clinical syndrome of familial tumor calcinosis. FGF-23 acts as a promising target of novel therapeutic interventions in chronic kidney disease and beyond, which should be tested in future clinical trials. The capture antibody is pre-coated on 96-well plates. The standards, test samples and biotin conjugated detection antibody are added to the wells subsequently, and washed with wash buffer. HRP-Streptavidin is added and unbound conjugates are washed away with wash buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of FGF23 in the sample.
