FGF2 also known as basic fibroblast growth factor belongs to the fibroblast growth factor (FGF) family, which plays an important role in the regulation of embryonic development, cell proliferation, and cell differentiation. FGF family members possess broad mitogenic and cell survival activities. FGF2 is present in basement membranes and in the subendothelial extracellular matrix of blood vessels. It stays membrane-bound as long as there is no signal peptide. FGF2 is a critical component of human embryonic stem cell culture medium and it is necessary for the cells to remain in an undifferentiated state, although the mechanisms by which it does this are poorly defined. FGF2 can induce angiogenesis and mediates phosphorylation of ERK1/2 and thereby promotes retinal lens fiber differentiation. Low levels of FGF2 play a key role in the incidence of excessive anxiety. FGF2 has a translational role in promoting adiposity-associated mammary epithelial cell transformation. FGF2, in conjunction with BMP4, promote differentiation of stem cells to mesodermal lineages. After differentiation, BMP4 and FGF2 treated cells generally produce higher amounts of osteogenic and chondrogenic differentiation than untreated stem cells. BioVision’s FGF2 (Human) ELISA Kit is used for the quantitative detection of FGF2 in serum, plasma, tissue homogenates and other biological fluids. It is based on the principle of sandwich ELISA. The capture antibody is pre-coated on 96-well plates. The standards, test samples and biotin conjugated detection antibody are added to the wells subsequently, and washed with wash buffer. HRP-Streptavidin is added and unbound conjugates are washed away with wash buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of FGF2 in the sample.
