Phagocytosis in mammals serves as an important first line defense mechanism against invading pathogens. It is also essential for continuous clearance of dying cells, tissue remodeling, and acquisition of nutrients for some cells. Phagocytosis is a specific form of endocytosis initiated by recognition and binding of foreign particles by cell surface receptors, followed by their engulfment, and formation of phagosomes. Maturing phagosomes transform to phagolysosomes which destroy the pathogen through enzymes and toxic peroxides. Zymosan prepared from yeast cell wall (Saccharomyces cerevisiae), and consisting of protein-carbohydrate complexes is frequently used as a pathogen in phagocytosis assays. BioVision’s EZCell™ Phagocytosis Assay Kit (Green Zymosan) utilizes pre-labeled Zymosan particles as a tool for rapid and accurate detection and quantification of in vitro phagocytosis by fluorescent microscope, spectrophotometer or flow cytometry. Our kit provides a robust screening system for activators and/or inhibitors of phagocytosis and Toll-like receptors ligands (TLR).

Can the kit work on bacteria or yeast cells?
The kit has been standardized for mammalian cells only
What is the exact volume of sample required for this assay?
There is no specific volume we can recommend for the amount any sample to be used since it is completely sample concentration and quality based. You have to do a pilot expt with multiple sample volumes to determine the optimal volume which gives a reading within the linear range of the standard curve. Please refer to the citations for this product to see what other clients have used with similar sample types.
Do you have trial sizes of this kit?

Can we purchase individual components of this kit?

Can we use an alternate buffer for sample preparation (cell lysis, sample dilutions etc)?
Our assay buffers are optimized for the reactions they are designed for. They not only contain some detergents for efficient lysis of your cells/tissue, but also contain some proprietary components required for the further reactions. Therefore, we highly recommend using the buffers provided in the kit for the best results.
When using neutrophils with K397-100, what is the best positive control?
Untreated neutrophils (not treated with inhibitors) are the best positive control for the assay. You may also use other phagocytic cells as positive control such as macrophages and PBMCs. Negative control will be wells without any cells.
Is there a good inhibitor of phagocytosis that can be used, not just a negative control?
You can use Cytochalasin D, Lantrunculin A etc.
Why do I need to quench my samples? At step VII-6, quenching is given as optional.
The quencher that we use will reduce the surface fluorescence/background fluorescence if it is high for your sample. It is optional because not all samples will give high background fluorescence and depending on your sample type, you can either opt for quenching or not.
