How are your fluorescent exosomes labeled? Is it a genetic approach or dye based labeling?
The exosomes are labeled post isolation. It is a dye-based labeling.
If you are using dye based labelling, is that a lipid dye or an antibody label? For how long is it stable?
It is lipid dye labeling. Exosomes can be aliquoted and stored at -20 °C for 6 months.
You use ultracentrifugation to prep the EVs from cell culture and bodily fluids for the non-fluorescent products. What is the method used to determine the size range? Do you also evaluate them using TEM?
We do not use ultracentrifugation anymore. The exosomes are purified by Tangential Flow Filtration (TFF) and size exclusion chromatography. The size is determined by Nanoparticle Tracking Analysis (NTA), using the Zetaview instrument of Particle Matrix. We do not evaluate all lots by Transmission Electron Microscopy (TEM), but we have done the testing. The vesicles are in size range 40 – 110 nm.
