Desmoglein 1 is a member of the desmosomal cadherin family that comprise the desmogleins and desmocollins. Desmosomes are cell-cell junctions between epithelial, myocardial, and certain other cell types. Desmoglein subfamily members have been identified and all are members of the cadherin cell adhesion molecule superfamily. Desmoglein-1 is the target antigen in majority of the cases linked to IgG/IgA pemphigus, which is an autoimmune IgG/IgA antibody mediated response. It is an essential intercellular adhesion molecule that is altered in various human cutaneous disorders. Desmoglein-1 is a target of Staphylococcus Exotoxins A and B which contribute to the pathoaetiology of Staph Scalded Skin Syndrome. Deficiency of the Desmoglein-1 protein has been found to be associated with increased expression of multiple genes encoding allergy-related cytokines. It is haploinsufficient and a mutation in the gene can cause the autosomal dominant mutation striate palmoplantar keratoderma. BioVision’s Desmoglein-1 (Human) ELISA Kit is used for the quantitative detection of Desmoglein-1 in serum, plasma, tissue homogenates and other biological fluids. It is based on the principle of sandwich ELISA. The capture antibody is pre-coated on 96-well plates. The standards, test samples and biotin conjugated detection antibody are added to the wells subsequently, and washed with wash buffer. HRP-Streptavidin is added and unbound conjugates are washed away with wash buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of Desmoglein-1 in the sample.
