D-Mannitol is one of the most abundant sugar alcohols in nature. It can be produced in the biosphere by a range of organisms such as bacteria, yeast, fungi and almost all plants. Mannitol functions as a carbohydrate storage depot, as a scavenger for reactive oxygen species and as an osmoprotectant. Recent studies have shown that increased mannitol content in plants protects them against fungal infections and improves resistance to drought and high salinity. The mannitol pathway reversibly converts fructose to mannitol and serves as the main carbohydrate cycle for fungi survival. Measurement of mannitol level is important for evaluating mannitol metabolic pathways and commercially, in developing fungi and drought resistant plants. In BioVision’s D-Mannitol Assay Kit, D-mannitol is converted to D-fructose by mannitol dehydrogenase in the presence of NAD to form NADH, which reduces a colorless probe to a chromogen with strong absorbance at 450 nm. L-Arabitol, another sugar alcohol, also acts as substrate for mannitol dehydrogenase & undergoes similar reaction; therefore, this kit can also be used to measure L-arabitol. The D-mannitol assay kit is fast (~15 minutes), sensitive & easy to use. This assay kit can detect mannitol level less than 1 nmol/reaction (< 10 µM) & can be used for a variety of sample types.

We would like to use this kit to measure mannitol in urine. Can this be done?
Urine samples after brief spinning down can be used directly in this assay. We suggest testing a few dilutions to ensure the values are within the linear range of the std. curve.
Is protein precipitation recommended for this kit?
No, protein removal is not necessary for this kit.
What endogenous molecules can potentially interfere in this assay?
NADH can interfere in this assay. Hence parallel background controls are to be run so that the background can be subtracted from the sample values.
Does the 100 assays include standards, controls and samples?
Yes, we provide sufficient reagents plus 10% in excess for 100 assays including stndards, samples and the necessary conrols.
Can this assay be normalized with respect to protein concentration?
Yes, a detergent-compatible BCA assay can be used for protein quantitation to normalize sample amount.
Can individual components of this kit be purchased separately?

Can alternate buffers be used for sample preparation (cell lysis, sample dilutions etc)?
Our assay buffers are optimized for the reactions they are designed for. They not only contain some detergents for efficient lysis of your cells/tissue, but also contain some proprietary components required for the further reactions. Therefore, we highly recommend using the buffers provided in the kit for the best results.
What is the shelf life of this kit?
This kit is good for 12 months from the date of shipment in the unopened form when stored at the appropriate temperature and appropriate conditions. After opening and reconstitution, some of the components in this kit are good for 2 months at -20°C. Please refer to the datasheet for storage information and shelf life of each of the components.
Does the slope of the standard curve in all experiments need to be the same? What is the acceptable variability?
The R squared value signifies the probability that the data points are on a straight line. As long as this value sis close to 98 or 99%, the std curve can be trusted. The slopes can vary depending on the instrument, the dilutions used etc.
