Ceruloplasmin is a copper containing protein found primarily in the blood. It carries approximately 70% of the total copper present in the blood. Ceruloplasmin exhibits a weak oxidase activity, which is a more accurate method of assessing ceruloplasmin in serum than immunodiffusion or other non-enzymatic assays. Normal Ceruloplasmin levels are generally 1-4 µM (15-60 mg/dl), equivalent to approximately 50-150 mU/ml. Elevated amounts of serum ceruloplasmin are found in pregnancy, cancer, rheumatoid arthritis and in several mental conditions such as Alzheimer’s, schizophrenia and OCD. Abnormally low amounts are found in Wilson’s and Menkes’ Diseases and in several other rare conditions. Has this kit been tested with sera from other species than human? Will it work with Chicken serum?
Since elevated amounts of serum ceruloplasmin are found in pregnancy and many human diseases like cancer, arthritis Alzheimer’s etc, we have tested this kit using human serum. The kit is optimized to be used with different mammalian sera. Theoretically it could work with chicken serum also. But since we have not tried this, there might be optimization necessary to get the best results.
Can serum samples from Wilson's disease mouse model be tested with this kit, especially since the ceruloplasmin levels are expected to be very low?
Our assay kit can measure ceruloplasmin activity in the range between approximately 5 and 500 mU/ml. If the expected range for the customer’s samples will fall in this range (typical ceruloplasmin levels for Willson’s disease is <0.2g/L), then our kit might work. The sample volume might need optimization to get the best results. As long as the mouse ceruloplasmin exhibits oxidase activity, it should be detected by this kit. What is the shelf-life of this kit before and after opening?
The shelf-life of the unopened K669 is 1 year at 4C. After opening the kit , for best results we recommend using up the components within 2 months.
Do you recommend any particular type of 96-well plate for this assay?
A flat-bottom clear polystyrene 96-well plate is what we use in-house.
How can the std. curve be plotted with Ceruloplasmin on the X axis?
You can use purified Ceruloplasmin in increasing amounts with this assay and plot OD versus amount in ug and then use this as the std. curve instead of the oxidized substrate.
Are trial sizes of this kit available?

Is it essential to make a standard curve for every expt, or is one curve/kit enough?
Yes, it is strongly recommended to do the standards every time you do the expt. There is always a chance that something was done differently that day and we do not want any conditions to differ between standards and samples.
