Details
Cathepsin E (CTSE, EC: 3.4.23.34) is a gastric aspartyl protease that functions as a disulfide-linked homodimer. This protease has a specificity similar to that of pepsin A and cathepsin D. It is an intracellular proteinase that is found in highest concentration on the surface of epithelial mucus-producing cells of the stomach. It is the first aspartic proteinase expressed in the fetal stomach and is found in more than half of gastric cancers. BioVision’s Cathepsin E Activity Assay Kit utilizes the ability of an active Cathepsin E to cleave a synthetic MCA-based peptide substrate to release free MCA, which can be easily quantified using a fluorometer or fluorescence microplate reader. This assay kit is simple, rapid and can detect Cathepsin E activity as low as 1 mU in biological samples.

Is the substrate cell permeant? Can this kit be used for immunofluorescence or FACS?
The substrate is not cell permeant and ths kit is not suitable for immunofluorescence or FACS based assay.
Could you let me know whether you have used baculovirus to produce this product?
None of the kit components have been made using baculovirus

Each kit lets you run 100 reactions, including all controls
Can the kit work on bacteria or yeast cells?
We have optimized the kit with mammalian samples. However, theoretically these kits should work with samples from multiple species/sources. Since the optimal conditions depend on the sample type, the protocol has to be be adapted to fit the samples for efficient results. Please refer to this kits citations to see what kind of samples have been used with this kit other than mammalian samples. The kit has been standardized for mammalian cells only.
What is the exact volume of sample required for this assay?
There is no specific volume we can recommend for the amount any sample to be used since it is completely sample concentration and quality based. You have to do a pilot expt with multiple sample volumes to determine the optimal volume which gives a reading within the linear range of the standard curve. Can we use an alternate buffer for sample preparation (cell lysis, sample dilutions etc)?
Our assay buffers are optimized for the reactions they are designed for. They not only contain some detergents for efficient lysis of your cells/tissue, but also contain some proprietary components required for the further reactions. Therefore, we highly recommend using the buffers provided in the kit for the best results.
