The branched-chain amino acids or BCAA’s, refer to the amino acids with non-linear aliphatic side-chains, namely leucine, isoleucine and valine. These three essential amino acids make up approximately 1/3 of skeletal muscle in the human body. BCAA’s are currently used clinically to aid in the recovery of burn victims, as well as for strength supplementation for athletes. BCAA’s, primarily Leu, can stimulate insulin secretion. The BCAA’s have also been implicated in a wide range of other physiological effects. BioVision's BCAA Assay Kit provides a simple convenient means of measuring the BCAA’s in a variety of biological samples. The kit utilizes an enzyme assay in which BCAA is oxidatively deaminated, producing NADH which reduces the probe, generating a colored product (λmax = 450 nm). BioVision’s BCAA kit measures BCAA’s in the range of 0 to 10 nmol per sample with a detection limit of ~0.2 nmol (~10 µM BCAA in sample). BCAA’s are present in serum ~ 0.1-0.4 mM each (~0.125-1.5 mM combined).

Does it detect only Leucine or total BCAAs?
K564 measures all Branched-chain amino acids. Leucine is one of them and hence is used as the standard. Any Leucine, Isoleucine and Valine in solution will be detected by this kit. If protein hydrolysis before the assay yields these amino acids, then the kit will detect them.
Can plasma samples be collected in EDTA tubes? Does the plasma need to be pre-treated with anything? Does the plasma need to be centrifuged? If so, at what speed and for how long and at what temperature?
Collect blood using heparin as the anticoagulant. EDTA will chelate metal iosn and is not advisable. Plasma needs no pretreatment. Centrifuge the blood at 1500-2000g for 10-20 mins at 4C to collect the plasma.
Can the kit work on bacteria or yeast cells?
The kit has been standardized for mammalian cells only.
What is the exact volume of sample required for this assay?
There is no specific volume we can recommend for the amount any sample to be used since it is completely sample concentration and quality based. You have to do a pilot expt with multiple sample volumes to determine the optimal volume which gives a reading within the linear range of the standard curve. What is the shelf life of this kit?
BioVisions’s assay kits expire 6 months from the date of shipment. Some components of the kit may expire sooner as mentioned in the data sheet

Can we use an alternate buffer for sample preparation (cell lysis, sample dilutions etc)?
Our assay buffers are optimized for the reactions they are designed for. They not only contain some detergents for efficient lysis of your cells/tissue, but also contain some proprietary components required for the further reactions. Therefore, we highly recommend using the buffers provided in the kit for the best results.
Could you please tell me whether the kit works on leucine derivatives such as L-leucyl-L-leucine?
The kit measures all branched chain amino acids. It cannot detect derivatives of leucine or derivatives of other branched chain amino acids.
