• Cross Reactivity: Avelumab infusion camouflages/masks the presence of antibody to Avelumab (ATP) in serum/plasma samples. Therefore, blood sampling time is critical for detection of antibodies. It is convenient to obtain blood sample just before the infusion or at least 2 weeks after the infusion of Avelumab.

Avelumab is a fully human anti-PD-L1 IgG1 lambda monoclonal antibody that has a molecular weight of approximately 147 kDa. Avelumab binds PD-L1 and blocks the interaction between PD-L1 and its receptors PD-1 and B7-1. By inhibiting PD-L1 interactions, avelumab is thought to enable the activation of T-cells and the adaptive immune system. By retaining Fc-region, avelumab is thought to engage the innate immune system and may induce antibody-dependent cell-mediated cytotoxicity. BioSim™ anti-Avelumab (Human) ELISA kit is designed to detect antibody against Avelumab with high specificity and sensitivity in biological matrices. It is based on the principle of sandwich ELISA. Controls and samples (serum or plasma) are incubated in the microtiter plate coated with the avelumab. After incubation, the wells are washed. Then, horse radish peroxidase (HRP) conjugated probe is added and binds to avelumab antibodies captured by the avelumab on the surface of the wells. Following incubation wells are washed and the bound enzymatic activity is detected by addition of tetramethylbenzidine (TMB) chromogen substrate. Finally, the reaction is terminated with an acidic stop solution. The color developed is proportional to the amount of avelumab antibodies in the sample or controls. The results can be evaluated with using cut-off value.
