What is the Mg2+ concentration in the 10X PCR buffer?
The concentration of Mg2+ in the 10X PCR buffer provided is 15 mM. The final concentration of Mg2+ in the reaction is 1.5 mM.
How can I optimize the PCR reaction for multiplexing?
The concentrations of DNA Polymerase, Mg2+ and the primers have to be optimized for each amplicon in multiplexing. The concentrations will need to be determined by the end user. The general rule would be to lower the concentrations of the primers for strongly amplified targets and increase the concentrations of the primers for weakly amplified targets.
