Adenosine, a purine nucleoside, is present throughout the body. It plays an important role in energy transfer via the formation of ATP, ADP and AMP and in signal transduction via the formation of cAMP. Adenosine mediates its effects directly via adenosine receptors A1, A2A, A2B and A3. It regulates myocardial oxygen consumption and coronary blood flow, exerts anti-inflammatory effects throughout the body and also regulates the Renin-Angiotensin system. It also plays a role in tissue damage and repair, and cell death. Plasma adenosine levels are increased in patients with ischemic and non-ischemic heart failure. In BioVision’s Adenosine Assay, adenosine is measured using adenosine deaminase followed by a multi-step enzymatic approach resulting in the generation of an intermediate that reacts with the Adenosine Probe with the formation of a fluorescent product. The fluorescent product is measured at Ex/Em = 535/587 nm. Detection range: 2-80 pmol.

Can ATP interfere with the assay?
The kit has a very specific converter system that helps in assaying for adenosine specifically as opposed to ATP, AMP and ADP.
Do you know what reagents can interfere with the assay?
Inosine, xanthine and hypoxanthine present in the sample(s) will contribute to the background. PBS, DMEM or RPMI should not contribute background as long as they do not contain supplemented inosine, xanthine/hypoxanthine.
Is it possible to use your adenosine kit to measure adenosine in a tissue sample?
We have not used this kit for tissue adenosine quantification. There might not be a large amount in this sample type to begin with, so you might need to optimize the sample amount needed. Homogenize tissues in the assay buffer in the kit and centrifuge to get a tissue lysate. Use this for the assay.
Can the kit work on bacteria or yeast cells?
The kit has been standardized for mammalian cells only
What is the exact volume of sample required for this assay?
There is no specific volume we can recommend for the amount any sample to be used since it is completely sample concentration and quality based. You have to do a pilot expt with multiple sample volumes to determine the optimal volume which gives a reading within the linear range of the standard curve. What is the shelf life of this kit?

Can we use an alternate buffer for sample preparation (cell lysis, sample dilutions etc)?
Our assay buffers are optimized for the reactions they are designed for. They not only contain some detergents for efficient lysis of your cells/tissue, but also contain some proprietary components required for the further reactions. Therefore, we highly recommend using the buffers provided in the kit for the best results.
