Acid phosphatase (AP) dephosphorylates phosphate groups from the phosphate esters in acid conditions. Different forms of acid phosphatase are found in different organs, and their serum levels are used as a diagnosis for disease in the corresponding organs. For example, elevated prostatic acid phosphatase levels may indicate the presence of prostate cancer and elevated tartrate resistant acid phosphatase levels may indicate the bone disease. BioVision’s Acid Phosphatase Fluorometric Assay Kit uses non-fluorescent Methylumbelliferyl phosphate disodium (MUP) as the substrate which has Ex/Em=360/440 nm when dephosphorylated by AP. The kit is an ultra-sensitive, simple, direct and HTS-ready fluorometric assay designed to measure AP activity in serum and other bio-samples. The detection sensitivity is ~1 µU, more sensitive than colorimetric assays. The kit is suitable for both research and drug discovery.

What is the source of the positive control?
The acid phosphatase in K421 is from wheat germ. 3-5 units per kit.
K421-500 and K422-500 both kit uses the same substrate MUP, then how are they specific?
" K421: Acid phosphatase(AP) can dephosphorylate MUP(4-Methylumbelliferyl phosphate disodium salt) to yield 4-methylumbelliferone, a fluorescent product, with Ex/Em at 364/448 nm (pH 10.3). The reaction can be stopped by 0.3M Glycine buffer. K422:Alkaline phosphatase(ALP) can dephosphorylate MUP(4-Methylumbelliferyl phosphate disodium salt) to yield 4-methylumbelliferone, a fluorescent product, with Ex/Em at 364/448 nm (pH 10.3). The reaction occurs in the presence of glycine. So having glycine as a component of the buffer, we eliminate AP activity and only measure ALP activity in K422."
Can the kit work on bacteria or yeast cells?
The kit has been standardized for mammalian cells only
What is the exact volume of sample required for this assay?
There is no specific volume we can recommend for the amount any sample to be used since it is completely sample concentration and quality based. You have to do a pilot expt with multiple sample volumes to determine the optimal volume which gives a reading within the linear range of the standard curve. Can we use an alternate buffer for sample preparation (cell lysis, sample dilutions etc)?
Our assay buffers are optimized for the reactions they are designed for. They not only contain some detergents for efficient lysis of your cells/tissue, but also contain some proprietary components required for the further reactions. Therefore, we highly recommend using the buffers provided in the kit for the best results.
