Acetylcholinesterase (AChE) is one of the most crucial enzymes associated with nerve response and function. AChE is a serine protease that hydrolyzes the neurotransmitter acetylcholine. Irreversible inhibition of AChE may lead to muscular paralysis, convulsions, bronchial constriction, and death by asphyxiation. BioVision’s Acetylcholinesterase Activity Assay Kit provides a simple and sensitive method for continuous monitoring of enzymatic activity using colorimetry (OD 570 nm). In this assay, AChE converts acetylcholine substrate to choline, which is then oxidized by choline oxidase (CO) to produce an intermediate. The intermediate reacts with a highly specific probe to generate color. This assay kit can detect acetylcholinesterase activity as low as 0.5 mU/ml in a variety of samples.

What is the source of the positive control?
The acetylcholine esterase is from Electrophorus electricus (Electric Eel).
Can I use this kit for measuring butyrylcholinesterase activity in plasma(or serum)?
The plasma BuCHE can hydrolyse many different choline esters but prefers butyryl choline or derivatives like it. This enzyme does not use acetyl choline as the preferred substrate but since this is a more generic esterase, it could use acetyl choline in absence of other substrates.
I am using recombinant protein with this kit. Can the presence of tag on the protein inhibit the reaciton?
Using recombinant acetylcholine esterase with tags should work as long as the tags do not affect the folding of the protein into its native active state.
Is DTT compatible with the assay?
According to the assay principle, Acetyl-cholinesterase converts acetylcholine substrate to choline, which is then oxidized by choline oxidase (CO) to produce an intermediate. The intermediate reacts with a highly specific probe to generate color. DTT and beta-mercaptoethanol should be avoided at concentration >10µM as the probe is not stable in their presence.
Can we use frozen samples with this assay?
We would recommend using fresh samples. However, frozen samples can be used too provided they have not undergone multiple freeze thaw cycles.
Can the kit work on bacteria or yeast cells?
The kit has been standardized for mammalian cells only.
What is the exact volume of sample required for this assay?
There is no specific volume we can recommend for the amount any sample to be used since it is completely sample concentration and quality based. You have to do a pilot expt with multiple sample volumes to determine the optimal volume which gives a reading within the linear range of the standard curve. Can we use an alternate buffer for sample preparation (cell lysis, sample dilutions etc)?
Our assay buffers are optimized for the reactions they are designed for. They not only contain some detergents for efficient lysis of your cells/tissue, but also contain some proprietary components required for the further reactions. Therefore, we highly recommend using the buffers provided in the kit for the best results.
