2-phosphoglycerate (2PG) is an important intermediate in the glycolysis pathway. 2-Phosphoglycerate is converted by enolase to phosphoenolpyruvate (PEP) which is a key step from glucose to pyruvate. Aberrant glycolytic metabolism is a highly studied and potentially critical mechanism for ATP generation in cancer cells (The Warburg effect). Measurement of intracellular 2PG levels is a useful tool for analyzing the glycolytic pathway and its relevance to cancer research. BioVision’s 2-Phosphoglycerate Assay kit is a sensitive, fast and easy-to-use kit. In this assay, 2PG is converted by Enzyme Mix to PEP, which is further converted to pyruvate. The pyruvate is oxidized to generate color (OD 570 nm) and fluorescence (Ex/Em = 535/587 nm). The colored product or fluorescence intensity is proportional to 2PG level. This assay kit can detect 2PG level below 20 pmol and can be used for a variety of sample types.

We checked a principle of the kit, and this kit will measure endogenous PEP and pryuvate in sample. The customer would like to measure only 2PG in sample, so this kit assumed to be unsuitable for his purpose.Is my understanding of this correct?
There is a misunderstanding. K778 is for measuring 2-Phosphogycerate (2-PG). In this assay, 2PG is converted by Enzyme Mix to PEP, which is further converted to pyruvate. The pyruvate is oxidized to generate color (OD 570 nm) and fluorescence (Ex/Em = 535/587 nm). The colored product or fluorescence intensity is proportional to 2PG level. So this kit IS SUITABLE for your customer's experiment.Endogenous PEP and pyruvate in the sample will also develop some color and hence a sample background control (without the enzyme mix) will take care of the PEP + pyruvate background. This value can be subtracted from the sample readings with enzyme mix to get the 2-PG concentration in the sample.
Can we use frozen samples with this assay?
We would recommend using fresh samples. However, frozen samples can be used too provided they have not undergone multiple freeze thaw cycles.
Can the kit work on bacteria or yeast cells?
The kit has been standardized for mammalian cells only.
What is the exact volume of sample required for this assay?
There is no specific volume we can recommend for the amount any sample to be used since it is completely sample concentration and quality based. You have to do a pilot expt with multiple sample volumes to determine the optimal volume which gives a reading within the linear range of the standard curve. Please refer to the citations for this product to see what other clients have used with similar sample types.

Can we use an alternate buffer for sample preparation (cell lysis, sample dilutions etc)?
Our assay buffers are optimized for the reactions they are designed for. They not only contain some detergents for efficient lysis of your cells/tissue, but also contain some proprietary components required for the further reactions. Therefore, we highly recommend using the buffers provided in the kit for the best results.
