We offer convenient kits for quantification of caspase activity in cell lysates. The assays utilize synthetic peptide substrates labeled with either AFC (fluorometric assay) or pNA (colorimetric assay). Activated caspases in apoptotic cells cleave the synthetic substrates to release free AFC or pNA, which is then quantified using fluorometer or spectrophotometer, respectively. Caspase assays are simple, straightforward, and can be adapted for 96-well plates.
Fluorometric and Colorimetric Assays of Caspase-3 Activity. Apoptosis was induced in Jurkat cells with camptothecin for 6 hours. Caspase assays were performed according to the kit manuals.
Jurkat cells were treated with Camptothecin (2 uM) for various hours. Caspase-3 fluorometric assay was performed according to the kit instructions. Results show that the optimum induction time is 4-8 hours.
