HMG-CoA Inhibitors

HMG-CoA reductase (or 3-hydroxy-3-methyl-glutaryl-CoA reductase or HMGCR) is the rate-limiting enzyme of the Mevalonate pathway, the metabolic pathway responsible for the biosynthesis of cholesterol. This enzyme has profound roles in lipid metabolism, glucose homeostasis and hence has long-reaching consequences in overall cardiovascular health, obesity and diabetes. This enzyme is thus the target of the widely used cholesterol-lowering drugs known collectively as the statins. HMGCR is also involved in the synthesis of sterols, isoprenoids and other lipids. This is why its medical significance has spread into various areas of human-disease-related research like inflammation, developmental abnormalities etc.

HMGCR inhibitors like statins have shown promise in treating a mouse model of multiple sclerosis, an inflammatory autoimmune disease. Inhibition of HMGCR activity and the lack of isoprenoids stemming from this inhibition, can lead to germ cell migration defects as well as intracerebral hemorrhage.
