Cell death or cytotoxicity is classically evaluated by the quantification of plasma membrane damage. Lactate dehydragenase (LDH) is a stable enzyme, present in all cell types, and rapidly released into the cell culture medium upon damage of the plasma membrane. LDH, therefore, is the most widely used marker in cytotoxicity study. BioVision offers two LDH Cytotoxicity Assay Kits for convenient quantification of LDH released from damaged cells. The first LDH-Cytotoxicity Assay Kit utilizes a coupled enzymatic reaction in which LDH oxidizes lactate to pyruvate which then reacts with tetrazolium salt INT to form formazan. The increase in the amount of formazan correlates directly to the increase in the number of lysed cells. LDH activity can be quantified at OD 500 nm. The new LDH-Cytotoxicity Assay Kit II utilizes a WST reagent in the enzymatic coupling reaction in which LDH oxidizes lactate to generate NADH, which then reacts with WST to generate yellow color. The assay is highly sensitive and thus regular culture medium containing 10% serum can directly be used in the assay. LDH activity can be easily quantified at OD 450 nm. Both LDH-cytotoxicity assays are easy-to-perform and applicable to a variety of cytotoxicity studies. Both assays take less than 1 hr.
